Statement of Need and

Objectives The purpose of this acquisition is to procure a Keyence BZ-X1000 All-in-One Fluorescence Microscope to support ongoing and future NHGRI research activities.

Objectives include: Enable high-resolution fluorescence and brightfield imaging of biological specimens, including fixed cells, live cells, and tissue sections from pre-clinical mouse models. Support visualization and quantification of gene expression, vector distribution, cellular pathology, and therapeutic response. Enhance experimental efficiency, reproducibility, and data quality through integrated imaging, automation, and analysis capabilities. Reduce dependency on shared imaging facilities and improve turnaround time for time-sensitive experiments. The system shall meet or exceed the following

requirements: All-in-one fluorescence and brightfield microscope system High-sensitivity fluorescence detection suitable for low-signal biological samples Capability to image fixed and live cells, as well as tissue sections Automated image acquisition and stage control Integrated quantitative image analysis software High-resolution imaging suitable for cellular and subcellular visualization User-friendly interface to enable operation by laboratory personnel Capability for multi-channel fluorescence imaging Data export and compatibility with standard analysis workflows Provide the Keyence BZ-X1000 All-in-One Fluorescence Microscope or equal system meeting all specified
